Description
Mouse fibula clamp for multiphoton microscopy
3D printable clamp holding 4 fibulae with medial surface upwards in a 35mm-dish. This was designed to be single extrusion, fused-filament 3D printed using bio compatible PETG, using a 0.35mm nozzle. We used this design in mitochondrial membrane potential, mitochondrial morphology and reactive oxygen species assays to immobilize freshly excised, cleaned live tibiae in upright, multiphoton microscopy time lapse recordings. Use it as disposable, submersible scaffold. Snap the base into a 35mm dish (sized for Olympus brand), add assay medium, add bones, and snap retainer in to hold bones in position. See related tissue handling instructions (for tibiae) in: https://www.protocols.io/view/preparation-and-shipping-of-mouse-tibiae-for-multi-14egn3e5ql5d/v1
The fibula uCT mesh is not to scale and was from a whole animal uCT mesh, a kind gift of Simon Melov, Buck Institute for Research on Aging. The clamp was designed based on this mesh.
